  • キニーネ・ショ糖混合刺激に対するクロキンバエ味覚毛の応答インパルス

    日本味と匂学会誌, 日本味と匂学会, 21, 3, 265-266, Dec. 2014, Peer-reviwed, クロキンバエふ節の味覚毛より記録したショ糖応答のインパルス頻度はキニーネにより抑制されると考えられてきた。しかしショ糖またはキニーネそれぞれの単独刺激に対する応答はそれぞれの濃度の増加で振幅減少を示し,先の頻度抑制は振幅の変化であり頻度の現象ではない可能性があった。そこで各条件についてインパルスの振幅を検討したところ,糖応答の抑制の起きる範囲では振幅の変化は僅かであり,その影響はほとんどないことが認められた。

Although it has been known that flies have specific bitter-sensitive taste cells in their contact chemosensilla, the mechanism by which those cells transduce the chemical signal into electrical activity has remained elusive. In this study, we first confirmed that type D4 and D5 tarsal chemosensilla of the blowfly Phormia regina responded well to bitter substances. Then, recording impulses from type D4 chemosensilla, we examined the possibility that a G-protein-coupled inositol 1,4,5-trisphosphate (IP(3))-dependent transduction cascade is of importance in the bitter-sensitive taste cells. We found that the response to bitter substances was depressed by specific inhibitors of G-protein, phospholipase C, or IP(3) receptor in the tarsal taste receptor cells. These results suggest that G-proteins mediate the IP(3) pathway in the transduction cascade in bitter-sensitive receptor cells. (C) 2009 Elsevier Inc. All rights reserved.

To study appetite modulation by odours, we established a new experimental system based on the modification of a previous method, where flies fed sucrose flavoured with D-limonene subsequently showed reduced appetite to plain sucrose. In this new experimental system, a fly population was divided into two groups: (1) the "simultaneous" group of flies was exposed to D-limonene and sucrose simultaneously for 10 min, and (2) the "separate" group of flies was exposed to sucrose alone for 10 min, and 1 h later, to limonene for 10 min. The appetite of flies in the "separate" group for sucrose was unaffected by the experiment, but the appetite of flies in the "simultaneous" group was significantly decreased, and this effect lasted for >= 3 days. To investigate if this appetite modulation by D-limonene was based on long-term memory formation (protein synthesis), we examined the effects of the protein synthesis inhibitor, cycloheximide. Injection of cycloheximide 1 h after exposure to limonene and sucrose inhibited the appetite suppression in the "simultaneous" group of flies. In addition, to exclude the possibility that D-limonene exerted its effect through taste, rather than odour, we examined the effect of removing of the olfactory organs, antennae and maxillary palps on appetite modulation by D-limonene. When the olfactory organs were removed, no reduction in appetite was observed in the flies in the "simultaneous" group, indicating olfaction. In our new and effective appetite-modulation system for flies, modulation of appetite by olfactory detection of D-limonene was shown. (C) 2008 Elsevier Ireland Ltd. All rights reserved.

    CHEMICAL SENSES, OXFORD UNIV PRESS, 31, 6, 497-504, Jul. 2006, Peer-reviwed, Flies provide a powerful model system for exploring signaling systems in gustatory receptor neurons (GRNs). To elucidate the cellular and molecular bases of these signaling systems, we sought to develop techniques to dissociate GRNs. We developed a primary culture of GRNs isolated from the labella of the blowfly, Phormia regina, 4-5 days after pupation. Dissected labella were treated with papain in a low Ca2+ saline solution and shaken in Leibovitzs L-15 medium supplemented with 20-hydroxyecdysone, L-ascorbic acid, and trehalose with a test tube mixer. Released cells were plated and kept at 29 degrees C in a medium containing fetal bovine serum. After a minimum of 2 days in culture, we observed survival or growth of bipolar cells with the characteristic morphology of GRNs. We also examined taste responsiveness by monitoring intracellular Ca2+ with a Ca2+-sensitive fluorescent dye, fluo-3. For some bipolar cells, application of sucrose, NaCl, or LiCl for 5-20 s transiently increased the intracellular Ca2+ levels in cell bodies for 20-30 s. The primary cell culture described here is useful for functional analysis of GRNs.

  • Preliminary analysis of the GABA-induced current in cultured CNS neurons of the cutworm moth, Spodoptera litura

    NEUROSCIENCE LETTERS, ELSEVIER IRELAND LTD, 381, 1-2, 125-130, Jun. 2005, Peer-reviwed, Properties of GABA-induced current in cultured CNS (ganglion) neurons of cutworm moths (Spodoptera litura) were studied using a whole-cell patch-clamp technique. CNS neurons ranging from 10 to 20 mu m in diameter were cultured for 4-7 days in MGM-464 medium. GABA-induced a current response in these neurons in a sigmoidally dose dependent manner where the Hill coefficient and EC50 were 2.2 and 33.0 mu M, respectively. The reversal potential of GABA-induced current was -2.5 mV, which is close to the CI- equilibrium potential that was calculated from chloride ion concentrations in the present experimental environment. Furthermore, the GABA-induced current response depended on the extracellular chloride ion concentration, indicating that the receptor regulates the CI- permeability of cells. The GABA-induced current was completely inhibited by the GABA(A) antagonist, SR95531, and activated by the GABAA agonist, muscimol but not by the GABA(B) agonist, baclofen. On the other hand, the GABAc agonist, CACA, also induced a little smaller current than the GABA-induced response. The pharmacological behaviors of the GABA-induced currents suggest that these cells contain GABA receptors that belong to the GABA receptor family including the Rdl GABA receptors identified in Drosophila neurons.   • G(q)alpha subunit mediates receptor site-specific adaptation in the sugar taste receptor cell of the blowfly, Phormia regina

The recent identification of G protein-coupled receptor proteins (GPCRs) has progressed molecular biological studies of gustatory systems, although the signal transduction mechanisms have not yet been fully elucidated. In vision and olfactory receptor cells, G(q) class G protein is known to be a major signal transducer. By functional blocking of intrinsic G(q) with an anti-G(q/11)alpha antibody, we investigated the roles of G(q) in the sugar receptor cell of the blowfly, Phormia regina. Before and after introduction of the anti-G(q/11)alpha antibody into the cell through the DOC-permeabilized cell membrane, we recorded the responses of the receptor cell to sucrose and D-fructose, which stimulate different receptor sites, respectively. The initial impulse frequency in response to either sucrose or D-fructose was not changed by antibody introduction, whereas the adaptation rate in sucrose stimulation, but not fructose stimulation, became slower after antibody introduction. These results indicate that: (1) G(q) is a regulator of adaptation in the sugar receptor cell of Phormia, rather than a transducer, and (2) different adaptation mechanisms are promoted by stimulations with sucrose and D-fructose. In the blowfly, Phormia regina, cGMP has been suggested to be a second messenger for the sugar receptor cell. Generally, cGMP is produced by membranous or soluble guanylyl cyclase (sGC), which can be activated by nitric oxide (NO). In the present paper, we electrophysiologically showed that an NO scavenger, 2-phenyl-4,4,5,5-tetramethylimidazoline-3-oxide-1-oxyl (PTIO), an NO donor, 1-hydroxy-2-oxo-3-(N-methyl-3-aminopropyl)-3-methyl-1-triazene (NOC 7) or an NO synthase (NOS) inhibitor, N-G-nitro-l-arginine methyl ester (l-NAME) specifically affected the response in the sugar receptor cell, but not in other receptor cells. PTIO, when introduced into the receptor cells in a sensillum aided by sodium deoxycholate (DOC, pH 7.2), depressed the response of sugar receptor cells to sucrose but did not affect those of the salt or water receptor cells. NOC 7, given extracellularly, latently induced the response of sugar receptor cells; and l-NAME, when introduced into the receptor cells, depressed the response of sugar receptor cells. The results clearly suggest that NO, which may be produced by intrinsic NOS in sugar receptor cells, participates in the transduction cascade of these cells in blowfly.

Three of the four chemoreceptor cells are called the sugar, the salt and the water receptor cells, respectively. However, the specificity of the remaining chemoreceptor cell, traditionally called the `fifth cell', has not yet been clarified. Referring to behavioral evaluation of the oral toxicity of monoterpenes, we measured the electrophysiological response of the `fifth cell' to these compounds. Of all the monoterpenes examined, d-limonene exhibited the strongest oral toxicity and induced the severest aversive behavior with vomiting and/or excretion in the fly. d-Limonene, when dispersed in an aqueous stimulus solution including dimethyl sulfoxide or an odorant-binding protein (OBP) found in the contact-chemoreceptor sensillum, the chemical sense-related lipophilic ligand-binding protein (CRLBP), evoked impulses from the `fifth cell'. Considering the relationship between the aversive effects of monoterpenes and the response of the `fifth cell' to these effects, we propose that the `fifth cell' is a warning cell that has been differentiated as a taste system for detecting and avoiding dangerous foods. Here we suggest that in the insect contact-chemoreceptor sensillum, CRLBP carries lipophilic members of the noxious taste substances to the `fifth cell' through the aqueous sensillum lymph. This insect OBP may functionally be analogous to the von Ebner's grand protein in taste organs of mammals.

  • Cyclic AMP diffusion coefficient in frog olfactory cilia

    BIOPHYSICAL JOURNAL, BIOPHYSICAL SOCIETY, 76, 5, 2861-2867, May 1999, Cyclic AMP (cAMP) is one of the intracellular messengers that mediate odorant signal transduction in vertebrate olfactory cilia. Therefore, the diffusion coefficient of cAMP in olfactory cilia is an important factor in the transduction of the odorous signal. We have employed the excised cilium preparation from the grass frog (Rana pipiens) to measure the cAMP diffusion coefficient. In this preparation an olfactory cilium is drawn into a patch pipette and a gigaseal is formed at the base of the cilium. Subsequently the cilium is excised, allowing bath cAMP to diffuse into the cilium and activate the cyclic nucleotide-gated channels on the plasma membrane. In order to estimate the cAMP diffusion coefficient, we analyzed the kinetics of the currents elicited by step changes in the bath cAMP concentration in the absence of cAMP hydrolysis. Under such conditions, the kinetics of the cAMP-activated currents has a simple dependence on the diffusion coefficient. From the analysis we have obtained a cAMP diffusion coefficient of 2.7 +/- 0.2 . 10(-6) cm(2) s(-1) for frog olfactory cilia. This Value is similar to the expected value in aqueous solution, suggesting that there are no significant diffusional barriers inside olfactory cilia. At cAMP concentrations higher than 5 mu M, diffusion slowed considerably, suggesting the presence of buffering by immobile cAMP binding sites. A plausible physiological function of such buffering sites would be to prolong the response of the cell to strong stimuli.

As an intracellular concentration of Cl-([Cl-](i)) is critically important for the proposed function, we tried to measure [Cl-](i) of the isolated newt olfactory receptors using chloride-sensitive fluorescent dye, N-(6-methoxyquinolyl)-acetoethyl ester (MQAE). We found that the average of Cl- concentration through the cell is about 40 mM. This value is fairly lower than 120 mM that was suggested from reversal potential of tail component of the olfactory response in the low Cl- bath solution. Because the reversal potential for the present [Cl-](i) is above the resting potential, opening of the Cl- channels may serve as a booster for the depolarizing odor-response.   • Filtering effect of cone oil droplets detected in the P-III response spectra of Japanese quail

We measured the P-III components of electroretinograms (ERG) from isolated retinas of Japanese quail. We recorded responses to monochromatic flashes of equal photon numbers, and found that the shape of the response spectrum is dependent on the incident direction of the flashes. The spectrum obtained with the flashes from the cornea side had a steeper peak around 500 nm than that with flashes from the receptor side. This is clear electrophysiological evidence of the filtering effect of the oil droplets in the cone cells, which has long been suspected. We analyzed these spectra with respect to the absorption spectra of cone pigments and transmittance spectra of oil droplets. (C) 1997 Elsevier Science Ltd.

We addressed this issue by comparing currents evoked by cAMP and IP3 in native and reconstituted membranes from olfactory cilia. We detected only a cyclic nucleotide-gated conductance in the native membrane but both cyclic nucleotide-gated and IP3-gated conductances in the reconstituted membrane. The magnitudes of the cyclic nucleotide- and IP3-gated conductances were not correlated with each other in reconstituted membranes, suggesting that cyclic nucleotide- and IP3-gated channels originate in different cellular compartments.

  • Cellular and molecular constituents of olfactory sensation in vertebrates

    Since the discovery of odorant-activated adenylate cyclase in the olfactory receptor cilia, research into the olfactory perception of vertebrates has rapidly expanded. Recent studies have shown how the odor discrimination starts at the receptor level: each of 700-1000 types of the olfactory neurons in the neural olfactory epithelium contains a single type of odor receptor protein. Although the receptors have relatively low specific affinities for odorants, excitation of different types of receptors forms an excitation pattern specific to each odorant in the glomerular layer of the olfactory bulb. It was demonstrated that adenosine 3',5'-cyclic monophosphate (cAMP) is very likely the sole second messenger for olfactory transduction. It was also demonstrated that the affinity of the cyclic nucleotide-gated channel for cAMP regulated by Ca2+/calmodulin is solely responsible for the adaptation of the cell. However, many other regulatory components were found in the transduction cascade. Regulated by Ca2+ and/or the protein-phosphorylation, many of them may serve for the adaptation of the cell, probably on a longer time scale. It may be important to consider the resensitization as a part of this adaptation, as well as to collect kinetic data of each reaction to gain further insight into the olfactory mechanism.
